چکیده

It has been noted in recent years that groups ofgenes are often used at multiple stages and multiple places of development. Comparison of the functions of each group of genes in different developmental context gave rise to the notion of "functional gene cassettes." These groups of genes can often be viewed as sets of tools; each group is suited to carry out a certain operation and may serve similar functions in several developmental processes. For example, neurogenic genes can be thought of as a set of tools for singling out cells from a group of equivalent cells. The affected cells could be neuroectodermal cells, myoblasts, or a number of other cell types encountered at various stages of development (see below). A corollary is that if one notices that one member of a group is used in any developmental process, then there is good reason to suspect that other members may also be used. This simple notion provides a rather useful way of attempting to identify genes that control a hitherto poorly characterized developmental process. In the following, we will illustrate this notion with examples of three groups of genes. All three groups have important functions in controlling early steps of Drosophila neurogenesis. Each group is also utilized in other developmental processes.

Integrase-directed recovery of functional genes from genomic libraries

Large population sizes, rapid growth and 3.8 billion years of evolution firmly establish microorganisms as a major source of the planet's biological and genetic diversity. However, up to 99% of the microorganisms in a given environment cannot be cultured. Culture-independent methods that directly access the genetic potential of an environmental sample can unveil new proteins with diverse functi...

Cassettes for PCR-mediated gene tagging in Candida albicans utilizing nourseothricin resistance.

In recent years a number of molecular tools have been reported for use in the human fungal pathogen Candida albicans, including PCR-mediated approaches for gene disruption, conditional expression and epitope tagging. Traditionally these methods have utilized auxotrophic markers; however, the availability of auxotrophic markers can be limiting and in some instances their use may also impact on t...

Prevalence of Class 1 Integrons and Extended Spectrum Beta Lactamases among Multi-Drug Resistant Escherichia coli Isolates from North of Iran

Background: Extended spectrum beta lactamases (ESBLs) are an important cause of transferable multidrug resistance (MDR) in gram-negative bacteria. The most described ESBL genes are generally found within integron-like structures as mobile genetic elements. The aim of this study was to identify the accompanying of class 1 integrons and ESBLs in the MDR E. coli isolates. Methods: Susceptibility t...

Design, simplified cloning, and in-silico analysis of multisite small interfering RNA-targeting cassettes

Multiple gene silencing is being required to target and tangle metabolic pathways in eukaryotes and researchers have to develop a subtle method for construction of RNA interference (RNAi) cassettes. Although, several vectors have been developed due to different screening and cloning strategies but still some potential limitations remain to be dissolved. Here, we worked out a simple cloning stra...

Development and validation of vectors containing multiple siRNA expression cassettes for maximizing the efficiency of gene silencing

BACKGROUND RNA interference (RNAi) was originally identified as a biological process in which short double-stranded RNA (dsRNA) suppress the expression of genes complimentary to the dsRNA. This cellular intrinsic gene silencing mechanism has subsequently been developed as a useful tool for studies of gene function. A major strategy for producing small interfering RNA (siRNA) in cultured cells i...

A second set of loxP marker cassettes for Cre-mediated multiple gene knockouts in budding yeast.

Heterologous markers are important tools required for the molecular dissection of gene function in many organisms, including Saccharomyces cerevisiae. Moreover, the presence of gene families and isoenzymes often makes it necessary to delete more than one gene. We recently introduced a new and efficient gene disruption cassette for repeated use in budding yeast, which combines the heterologous d...
